My post (3 years ago) — still valid today!!! Academic system is built in a way that is intrinsically wrong and unjust — if you don’t get tenure-track faculty position after being postdoc and maybe later, instructor — it would be a time to change occupation. Each faculty position has dozens and even hundreds of candidates applying for it. Here in Israel, there is “glass ceiling” you can not pass through it. They don’t take regular candidates from outside world. System does not accept outsiders. After all the effort for PhD and postdoc, the whole biology business is just stupid waste of time and money. I would not recommend my kids career in biology with all my passion for science and research work. These guys, PIs think only about themselves, as mentors they are complete failure. Just in case, someone would ask my opinion on biology business…

Comment regarding “dominant-negative phenotype” of the mutant with disrupted RING Finger domain in E3 Ligase (Reference: eLife 2022;11:e71928).

Very interesting mechanism to account for the phenotype of the mutant disrupting RING Finger E3 Ligase Mib1! A mutant E3 Ligase binds, traps ubiquitin conjugation substrate proteins but fails to ubiqutinate them, functions like a “molecular sponge” — common feature of so-called “dominant-negative mutants”. There are other paradigms in Dev Biology (besides Notch/Delta and Wnt Signaling), in which such phenomenon has been noted — for example, Sobko A, Ma H, Firtel R, Dev Cell, 2002. It is very exciting that the authors engineered RING Finger mutant using CRISPR-Cas9 Gene Editing.
